Carbonic anhydrases (CAs) are essential modulators of the bicarbonate and proton concentrations, mediating the cell pH. This modulation is critical for cell survival and proliferation. The sulfonamide-based inhibitors and monoclonal antibodies inhibit the activity of [CA proteins IX]( and [XII]( in solid tumors. These inhibitors and antibodies are showing promising results in cancer clinical trials, including a combination trial of CA IX inhibitor with Nivolumab starting this year at Memorial Sloan Kettering Cancer Center.
All the other catalytically active isoforms of CA (I, [II]( [III]( [IV]( CAs [VA]( CAs VI-VII, CAs [XIII]( and [XIV]( are also found to be dysregulated in various types of cancer. They could potentially serve as prognostic and diagnostic markers in the near future following further exploration.
