Interleukin-18 ([IL-18]( is a pleiotropic pro-inflammatory cytokine. It belongs to the [IL-1 superfamily]( and participates in innate and acquired immunity. Recent studies have implicated the IL-18 pathway in autoimmune diseases, metabolic disorders, and cancers.
IL-18 mediates host immunity by regulating [IFN-γ]( production in synergy with [IL-12](. In the presence of [IL-2]( IL-18 stimulates natural killer (NK) cells, NKT cells, and established Th1 cells to produce [IL-3]( [IL-9]( and [IL-13](. IL-18 similarly activates downstream signal transduction as [IL-1β](. IL18 binds with its receptor [IL-18 receptor α (IL-18Rα)](. [IL-18Rβ / IL-18RAcP]( is subsequently recruited to the complex, and the downstream cellular signal is initiated. IL-18 signaling is negatively regulated by the secreted antagonist receptor [IL-18BP](. IL-18Rα may also bind [IL-37]( in association with [SIGIRR]( to transduce an anti-inflammatory signal.
