[Register Today!]( Streamlined for Speed: NEBNext UltraExpress™ for DNA library prep Thursday, January 25th 1:00 PM EST As next generation sequencing technologies improve and capacities expand, library construction has increasingly become the bottleneck for faster data turnaround and overall cost. There is a growing need for faster, streamlined, and automatable protocols that perform reliably without compromising the data quality of the libraries. To address this need, we developed two new single-tube “DNA sample in, library out” workflows with either mechanical (NEBNext UltraExpress™ DNA) or enzymatic (NEBNext UltraExpress™ FS DNA) shearing approaches to accommodate multiple applications. These robust and streamlined workflows have a one-size-fits-all protocol for express library preparation from a variety of sample types and input amounts (10 ng to 200 ng). These workflows reduce hands-on and total incubation time to less than two hours without sacrificing library quality or sequencing metrics. Furthermore, all steps in the workflow have been optimized to make them automation friendly. During this webinar, we will describe the NEBNext UltraExpress™ DNA and NEBNext UltraExpress™ FS DNA library preparation workflows. We will also discuss how these workflows efficiently generate high quality sequencing libraries across different animal, microbial, and plant species. We expect these library preparation methods to serve in a myriad of NGS applications, including clinical and diagnostic settings. Karen McKay Development Associate Karen joined NEB in 2015 after a previously successful and rewarding career working in the field of human in vitro fertilization as a senior embryologist. Her interest in preimplantation genetic testing led her to her current role as a Development Associate within the NEBNext® NGS product development group. During the past 8 years, Karen has contributed to the development of several key NEBNext products, including NEBNext Ultra II FS DNA (PCR and PCR-free) Library Prep Kits, Multiplex Oligos (UDI Adaptors and Primers) for Illumina, and most recently, the NEBNext UltraExpress DNA Library Prep Kit. Karen enjoys interacting with customers and has participated in many NEBNext hands-on training courses in the US. Kaylinnette Pinet Development Scientist Kaylinnette’s experience with wet lab research throughout her undergraduate and graduate studies yielded a great appreciation for quality reagents and reliable protocols. After earning a doctoral degree from Tufts University, her interest in molecular biology and novel biotechnology led her to NEB, initially as a postdoctoral researcher and then as a development scientist within NEBNext’s product development group. The NEBNext product line supports researchers by providing high quality reagents for both DNA and RNA sample prep, as well as target enrichment of DNA for NGS. Kaylinnette has helped develop several NEBNext® NGS products, including the NEBNext UltraExpress™ FS DNA Library Prep Kit. --------------------------------------------------------------- Streamlined for Speed: NEBNext UltraExpress™ for RNA library prep Tuesday, January 30th 1:00 PM EST RNA sequencing (RNA-seq) has become an invaluable tool in the study of biology, but it often requires multiple days to process samples from total RNA to final libraries. When working with samples in a high-throughput capacity in particular, protocol transfer to automation and the time required to prepare libraries can be a great hindrance to processing samples efficiently. We’ve developed a new protocol that allows users to easily generate robust RNA-seq libraries from a variety of sample types in a single day from 25-250 ng total RNA. Compatible with poly(A) enrichment or ribosomal RNA (rRNA) depletion, this new workflow has reduced preparation time by about 40% compared to leading library preparation kits, without sacrificing quality of libraries or sequencing metrics. In addition to faster preparation times, we introduced flexibility for users with a protocol that is automation friendly and allows users to implement a single protocol for all RNA inputs within the kit range. In addition to master mixed components and reduced incubation times, the reduction in bead cleanup steps significantly reduces hands-on time for library preparation, simplifying and streamlining the process. Furthermore, all steps in the workflow have been optimized to make them automation friendly.During this webinar, we will describe how the NEBNext UltraExpress™ RNA library preparation workflow provides a great advantage in efficiently generating strand specific/directional RNA-seq libraries while simultaneously maintaining high quality sequencing results, including 5’-3’ transcript coverage, excellent transcript abundance detection, and high correlation in transcript expression between replicates and across inputs. We expect these library preparation methods will serve in a myriad of NGS applications, including clinical and diagnostic settings. Chen Song Development Scientist Chen is a development scientist at NEB focusing on developing NGS workflows for DNA and RNA sequencing and is passionate about its applications in molecular diagnosis of cancer, infectious disease, and autoimmune disease. Prior to joining NEB, Chen had 10 years of research experience in assay development for biomarker discovery and therapeutics. During the past 8 years, Chen has contributed to the development of several key NEBNext® products, including the Immune-Seq kit, Multiplex Oligos (UDI Adaptors and Primers) for Illumina, and most recently, the NEBNext UltraExpress DNA Library Prep Kit.
