Pau D’Arco bark contains a number of compounds that have interesting biological effects. The Pau D’Arco compound that has garnered most interest is called beta-lapachone. In these experiments, the researchers evaluated the effect of beta-lapachone on arthritis and inflammation. “The purpose of this study was to evaluate the anti-inflammatory and anti-arthritic activities of beta-lapachone and to elucidate its probable mode of action.” To do this, they set up a couple of experiments with mice. In one of the experiments, they exposed mice to an inflammatory substance called carrageenan by injecting it into the paws of the animals. Carrageenan causes inflammation just like that seen in arthritis. “Carrageenan-induced paw edema, cell migration evaluation, and production of pro-inflammatory cytokines were used for this study.” The second series of experiments used chemicals to stimulate the process of arthritis. “Freund’s complete adjuvant (FCA)-induced arthritis was used as a model of chronic inflammation.” In these experiments, they exposed some of the mice to those inflammatory chemicals. And of those mice, some were given beta-lapachone. The researchers tested the effects of different oral doses of beta-lapachone supplement on inflammation and arthritis. “Beta-lapachone was tested in doses of 40 mg per kilogram and 60 mg per kilogram.” Animals given the high dose of beta-lapachone had far less swelling (edema) in response to the inflammatory substances. “In the paw edema test, the dose of 60 mg/kg gave a higher percentage inhibition of edema (49.3 %) than control.” The researchers tested inflammation by looking at a number of inflammatory markers: tumor necrosis factor (TNF-α), interleukin (IL-6), and nitric oxide (NO). Those tests showed that beta-lapachone lowers inflammation. “Beta-lapachone reduced concentrations of TNF-α, IL-6, and NO.” Neutrophils are a type of white blood cell. In rheumatoid arthritis, neutrophils begin to attack the joints - causing inflammation and arthritis. The researchers found that beta-lapachone decreased the attacks of neutrophils on body tissues. And that does mean it could be effective against rheumatoid arthritis. “Beta-lapachone inhibited neutrophil white blood cell migration.” The herbal compound was effective at lowering swelling in the arthritis test too. “In the arthritis test, beta-lapachone inhibited edema.” All of the tests showed positive results. The experiments indicated that beta-lapachone and Pau D’Arco could be beneficial for a number of different types of arthritis. “Significant anti-inflammatory and anti-arthritic activities were observed in animals treated with beta lapachone.” The remedy lowers inflammation, a problem in all types of arthritis. Beta lapachone also calms the immune system, a problem specific to rheumatoid arthritis. “The effects of beta-lapachone can be attributed in part to immunomodulation with reduction of pro-inflammatory cytokines.” The treatment also lowered nitric oxide, which is elevated in people with inflammatory diseases such as arthritis. “Beta-lapachone reduced serum concentrations of nitric oxide, an important mediator in the pathogenesis of rheumatoid arthritis.” The experiments show that herbal remedies such as Pau D’Arco, which contain beta-lapachone, could have numerous beneficial effects on arthritis.
